FIELD: biotechnology.
SUBSTANCE: disclosed is recombinant plasmid DNA phPGK-eGFP-CATTCGCC-CMV-mCherry, providing expression of reporter protein eGFP in cultured cells of Chinese hamster ovary, size 6,214 bps, having nucleotide sequence SEQ ID NO 2. Said plasmid contains the following elements in accordance with the graphic map presented in Fig. 1: human hPGK gene promoter and a transcription terminator of 182 bps, presented in SEQ ID NO 1 and consisting of 3'-inverted terminal repeat of piggyBac transposon (3'-TR), a polyA polyadenylation signal and having a motif CATTCGCC; eGFP gene sequence flanked by unique SmaI and EcoRI restriction sites; sequence of red fluorescent protein mCherry under control of CMV promoter and SV40 transcription terminator; ampicillin resistance gene AmpR, required for selection of target plasmids at the cloning step, ori and f1 ori plasmid replication origins.
EFFECT: invention provides high expression of the eGFP reporter protein in cultured Chinese hamster ovary cells.
