FIELD: biotechnology.
SUBSTANCE: group of inventions relates to sequencing of nucleic acids, specifically to a flow cell, a method of forming a cationic polymer hydrogel and a kit for introducing positive charges into a starting polymer hydrogel in a flow cell for sequencing. Flow cell comprises: a substrate, a cationic polymer hydrogel on the substrate and an amplification primer attached to the cationic polymer hydrogel. Cationic polymer hydrogel includes a cationic functional group, i) integrated into the monomer unit of the starting polymer hydrogel or ii) attached to the monomer unit of the starting polymer hydrogel by means of a linker. Method of forming a cationic polymer hydrogel includes introducing a fluid medium comprising a positively charged functional group, into a flow cell and incubating the initial polymer hydrogel in a fluid medium to form a cationic polymer hydrogel comprising a cationic functional group.
EFFECT: group of inventions reduces the probability of accidental binding of library fragments on the surface of the flow cell and reduces or prevents the seeding of the library fragments on the side walls of the flow cell, which increases the efficiency of library seeding.
