SUBSTANCE: invention relates to enzymology of single molecules and can be used to study enzymatic activities of single enzyme molecules using nanotechnology, as well as processes of denaturation of single enzyme molecules. Sequencing kit includes: a flow cell comprising: a substrate; catalytic polymer hydrogel on a substrate; an amplification primer attached to a catalytic polymer hydrogel; cleavage mixture containing a component which initiates cleavage of the blocking group. Catalytic polymer hydrogel includes a deblocking catalyst which accelerates cleavage of 3' OH blocking group of a blocked nucleotide introduced into a flow cell and embedded in a matrix chain attached to an amplification primer. Method of sequencing in a flow cell includes introducing the added mixture into the flow cell; embedding the individual nucleotides into corresponding nascent chains along the template chains; removing the added mixture; inclusion of individual nucleotides using optical imaging; and introducing the splitting mixture.
EFFECT: group of inventions provides improved release kinetics.
