FIELD: molecular biology.

SUBSTANCE: described is a method for filling reaction wells of a microfluidic chip of a disposable cartridge for nucleic acid amplification. In the disclosed method, the actuator closes the vent hole of the chip and creates a vacuum inside the channels with cells through the loading hole using the pump included in the actuator or the cartridge slide. Further, the actuating device supplies a reaction mixture to the loading opening by means of a disposable cartridge, which completely fills the channels and reaction wells, wherein the pressure inside the channels and the wells becomes equal to the atmospheric pressure.

EFFECT: preventing mixing of the contents of these wells both during their filling and during the nucleic acid amplification process therein.
