FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and represents a method of producing phycobiliproteins, namely natural pigments of C-phycoerythrin and C-phycocyanin, extracted from cyanobacterium Leptolyngbya cf. ectocarpi. A method of producing phycobiliproteins from cyanobacteria Leptolyngbya cf. ectocarpi provides for obtaining biomass with high content of phycoerythrin and phycocyanin by culturing cyanobacteria on a modified nutrient medium BG11 (g/l): NaNO3 – 3.37; NaH2PO4 × 2H2O – 0.065; cіtric acid – 0.006; Na2EDTA – 0.001; FeC6H5O7 – 0.006; 1 ml of a solution of trace elements of the following composition (g/l): H3BO3 – 2.86; CuSO4 × 5H2O – 0.08; ZnSO4 × 7H2O – 0.22; CoCl2 × 6H2O – 0.05; MnCl2 × 4H2O – 1.81; NaMoO4 × 2H2O – 0.4; prepared on sterile sea water for 10 days in plane-parallel photobioreactors with a working layer of 5 cm and volume of 10 l to density of 16 g of raw biomass per 1 litre of medium under 24-hour illumination of 13 klx and temperature of 34 °C with subsequent lyophilization of cyanobacteria biomass and extraction of phycobiliproteins from lyophilized biomass with distilled water at temperature of 38 °C (1:1 lyophilized cyanobacteria biomass and distilled water) and lyophilisation of phycobiliprotein extract.
EFFECT: invention enables to extract phycobiliproteins – phycoerythrin and phycocyanin – from biomass of cyanobacteria Leptolyngbya confer ectocarpi using a single-step method, which is promising for further use on an industrial scale.
