FIELD: analysis of biomaterials using flow cytometry.
SUBSTANCE: invention relates to a method for analyzing the effectiveness of phagocytosis. Method involves phagocytosis using phagocytosis objects pre-conjugated with a fluorescent label and biotin, and a sample of cells capable of phagocytosis. After phagocytosis is stopped, a sample of phagocytosed cells is stained with streptavidin conjugated with a fluorescent label. Then, using a flow cytometer with visualization, the number and spatial arrangement of phagocytosis objects are analyzed, where the number of phagocytosis objects and their arrangement is one or more fluorescence measurements, which are grouped by gating fluorescence intensity. Emission spectrum of the fluorescent label conjugated with the phagocytosis object does not overlap with the emission spectrum of the fluorescent label conjugated with streptavidin, and both spectra are in the detection range of the flow cytometer.
EFFECT: high accuracy of determining localization of phagocytosis objects inside a cell.
