FIELD: biotechnology.
SUBSTANCE: invention relates to analysis of biomaterials using flow cytometry and a method of sorting phagocytic cells. Method involves phagocytosis using phagocytosis objects pre-conjugated with a fluorescent label and biotin, and a sample of cells capable of phagocytosis. After phagocytosis is stopped, a sample of phagocytosed cells is stained with streptavidin conjugated with a fluorescent label. Then, using a flow cytometer with visualization and possibility of sorting, cells in the sample are sorted according to the number and spatial arrangement of phagocytosis objects, where the number of phagocytosis objects and their location inside or outside the cell is one or more fluorescence measurements, which are grouped by gating fluorescence intensity. Emission spectrum of the fluorescent label conjugated with the phagocytosis object does not overlap with the emission spectrum of the fluorescent label conjugated with streptavidin, and both spectra are within the detection range of the flow cytometer.
EFFECT: high accuracy of determining localization of phagocytosis objects inside a cell and sorting them.
