SUBSTANCE: described is a method for genotype-specific amplification of rotavirus cDNA by multiplex polymerase chain reaction. In the method, which includes the extraction of viral RNA, the production of cDNA, which is used as a matrix when performing PCR with electrophoretic visualization of amplification products in agarose gel, a series of PCR is carried out using oligonucleotides SEQ ID NO 1-21 to form reaction mixtures in which oligonucleotides SEQ ID NO 1-21 are distributed in the reaction mixtures as follows to detect: VP1 gene: SEQ ID NO 1, SEQ ID NO 2, SEQ ID NO 3; VP2 gene: SEQ ID NO 4, SEQ ID NO 5, SEQ ID NO 6; VP3 gene: SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9; NSP1 gene: SEQ ID NO 10, SEQ ID NO 11, SEQ ID NO 12; NSP2 gene: SEQ ID NO 13, SEQ ID NO 14, SEQ ID NO 15; NSP3 gene: SEQ ID NO 16, SEQ ID NO 17, SEQ ID NO 18; NSP5/6 gene: SEQ ID NO 19, SEQ ID NO 20, SEQ ID NO 21.
EFFECT: invention due to selected primers for multiplex PCR allows not only to differentially identify rotaviruses of the first and second genogroups, but also to detect intergenogroup reassortants in case, when reassortment has affected the least studied segments, in order to monitor circulation of rotaviruses and study their prevalence and variability.
