FIELD: biotechnology.
SUBSTANCE: invention relates to molecular biology and virology, specifically to a method for preparing CCHF virus cDNA samples recovered from biomaterial samples and virus-containing culture fluid, to sequencing a full-length genome sequence of the virus using different platforms. Summary of invention consists in use of developed set of oligonucleotide primers for specific enrichment of genomic cDNA of CCHF virus by PCR method in format of multiplex using 12 pools of oligonucleotide primers and monoplex PCR using 33 pairs of oligonucleotide primers. Invention enables to amplify the full-length genomic cDNA of the CCHF virus in form of 33 overlapping short fragments (with an average length of 750 bp), which implies the possibility of enrichment of fragmented cDNA contained in samples of clinical and field material. Enrichment of the genomic cDNA using the developed panel of primers can be carried out in the format of multiplexed PCR and monoplexed PCR.
EFFECT: obtained amplicons can be sequenced using different platforms for high-throughput and capillary sequencing.
