SUBSTANCE: disclosed is a nutrient media composition for culturing CHO cells expressing the monoclonal antibody pembrolizumab, which contains BalanCD HEK293, CDM4HEK293, nutritional supplement Feed BalanCD HEK293, 200 mM GlutaMax and water for injections. Said composition can be used in a method of producing a protein of interest. During the cultivation process, the carbon dioxide supply in the incubator is maintained at level of 6 %, the humidity at level of 85 %, orbital shaker torsion level of 120 rpm; feeding a nutritional additive Feed BalanCD HEK293 to maintain the CHO cell culture on 3rd, 7th, 9th, 11th and 13th day of cultivation in amount of 5 % of the volume of the culture fluid; reducing the initial temperature in incubator from 37 to 32 °C on the fourth day when the cell density is higher than 6×106 cells/ml, glucosamine hydrochloride is added on 8th day of cultivation.

EFFECT: use of said composition of nutrient media increases production of protein of interest.
