FIELD: chemistry.

SUBSTANCE: invention relates to a microfluidic cartridge for high amplification of polynucleotide-containing samples. Technical result is achieved by a microfluidic cartridge having a front side and an opposite back side. Cartridge comprises a first amplification chamber, a second amplification chamber, a first inlet located on the front side and in fluid communication with the first amplification chamber, a second inlet located on the front side and in fluid communication with the second amplification chamber, and a compressible contact pad glued to the front side. Compressible contact pad is configured to provide more uniform contact between the microfluidic cartridge and the plurality of contact heat sources, thereby providing more complete and consistent heat transfer to the first amplification chamber and a second amplification chamber from a plurality of contact heat sources in contact with the back side of the microfluidic cartridge. Compressible contact pad includes a first window above the first amplification chamber and a second window above the second amplification chamber. First window and the second window are configured to transmit light through the front side of the microfluidic cartridge to and from the first amplification chamber and the second amplification chamber, respectively.

EFFECT: improved thermal homogeneity and efficiency of amplification in the cartridge; the analyses demonstrate an improved limit of detection and an improved limit of quantitative determination.
