FIELD: microfluidics; biotechnology; medicine.
SUBSTANCE: present invention relates to the field of microfluidics, biotechnology, and medicine; it can be used for express PCR diagnostics, in particular for establishing the fact of patient’s infection and quality control of food products in direct proximity to a point of care diagnostics and ambulatory by detection of pathogens of Enterobacteriaceae family. The platform contains microfluid chambers connected by microfluid channels with valves located in them. Microfluid chambers, channels, and valves are made with flexible membranes providing the possibility of control of the movement of fluids due to the deformation of the specified flexible membranes, when they are pressed by an operator’s finger. Detection of the presence of biological pathogens in the sample is confirmed visually by change in color of the solution. The microfluid platform provides four stages of analysis – isolation of DNA, purification of DNA, amplification, and optical analysis in a single test system based on a disposable microfluid cartridge.
EFFECT: system is easy and reliable in implementation and provides the results during less than 3 hours after taking a sample; it can be used both in clinical laboratories and in field conditions of mobile centers; it will allow for increase in the reliability and biological safety of PCR test, reduction in the cost of analysis.
