METHOD OF TUBERCULIN PREPARING Russian patent published in 1998 - IPC

FIELD: veterinary science, microbiology. SUBSTANCE: tuberculin is obtained by culturing tuberculosis mycobacteria on synthetic nutrient medium and cultural fluid is separated. Tuberculin is isolated from cultural fluid by its treatment with trichloroacetic acid followed by reprecipitation with ammonium sulfate and desalting. Desalted concentrate is fractionated on ultrafilter membranes at "НОММ" index 150-1 000 kDa. High-molecular fraction is diafiltered on the same membranes. Then obtained permeates are subjected for sterilizing microfiltration. Sterile protein solution can be used for preparing dry "ППД"-tuberculin or "ППД"-tuberculin in standard solution. Method ensures to obtain highly specific, nonimmunogenic preparation for allergy diagnosis in agriculture animals. EFFECT: improved technology of preparing, increased yield of tuberculin. 5 tbl
