FIELD: microbiology, genetic engineering, molecular biology. SUBSTANCE: method involves transformation of the strain E.coli SG 200-50 with recombinant plasmid pLT21 with DNA fragment encoding human lymphotoxin. Cell are cultured, primary transformants are selected on medium with ampicillin. For amplification with plasmid DNA the repeated sowing of selected clones is carried out on nutrient medium containing, except for ampicillin, chloramphenicol at concentration 150 mcg/ml. Flasks are sown with liquid (+ ampicillin) medium obtained by washing out colonies grown at preliminary stage. Biomass is gathered at the end of logarithmic growth phase of culture.
