Abstract RU 2679076 C1

FIELD: biotechnology.

SUBSTANCE: group of inventions relates to the field of biotechnology, genetic engineering and medicine. Proposed artificial gene YkuJ-MPER encodes a protein-immunogen YkuJ-MPER against HIV-1 with the nucleotide sequence of SEQ ID NO:1 of length of 360 bps. Recombinant plasmid DNA of pET21a-YkuJ-MPER type is also proposed, designed to produce a protein-immunogen that induces HIV-specific B-cell response, with a size of 5,700 bps and a molecular weight of 3.8·103 kDa containing the YkuJ-MPER gene, encoding the YkuJ-MPER anti-HIV-1 protein-immunogen, under the control of the RNA polymerase promoter of phage T7, ensuring its expression in E. coli cells. Artificial YkuJ-MPER immunogen protein is also proposed, consisting of a B. subtilis YkuJ protein site and two HIV-1 MPER sites, having a molecular weight of 14.2 kDa and the amino acid sequence of SEQ ID NO:2.

EFFECT: group of inventions provides an effective induction of broadly neutralizing anti-HIV-1 antibodies in the selection of the optimal immunization strategy.
