SUBSTANCE: method provides DNA recovery from biological material samples, polymerase chain reaction with recovered DNA and amplification product detection. Target amplicon detection as PCR products testifies to the presence of Cryphonectria parasitica in the analysed sample. DNA recovery from vegetable tissue samples is ensured by using lytic buffers containing antioxidants, while the PCR requires applying the primers specific to low-polymorphous sequences of precursor protein genes of C.parasitica sex pheromones.
EFFECT: invention allows recognising efficiently a pathogen and can be applied for quick diagnosing of chestnut blight.
