SUBSTANCE: method for attenuation of the vaccine strain Francisella tularensis subsp.holarctica 15/10 provides the introduction of gene recD deletion in chromosomal DNA F. tularensis 15/10 with the help of the suicide plasmid pPVΔrecD. The suicide plasmid used for attenuation of the vaccine strain is produced on the basis of the plasmid pUC19 with the genes sacB, cat, mob and a fragment of chromosomal DNA F. tularensis 15/10 with gene recD deletion. A method for attenuation of the strain F. tularensis. subsp.holarctica 15/10 under the invention leads to producing the strain F. tularensis 15/10ΔrecD with gene recD deletion in a low-virulence genome preserving genome persistence and protective properties for the vaccine strain.

EFFECT: strain is applicable for preparing the tularemia live vaccine.
