SUBSTANCE: antitemplate plasmid vector consists of selection genetic marker with promoter and terminator, point of replication initiation and polylinker, containing 15 or more unique hexanucleotidic cleavage sites by restriction endonucleases, 3 or more octanucleotidic cleavage sites by restriction endonucleases and two recognition sites Xcml, located in the centre of polylinker area in reversed orientation and forming odd protruding single residues of deoxythymidine on 3 ends of DNA chains upon splitting by the restrictase. Also linear form of antitemplate vector is proposed. Also a set containing antitemplate plasmid vector or antitemplate liner vector, lygase and the set application sheet are proposed.

EFFECT: invention allows obtaining vectors providing capability of direct cloning of polymerase chain reaction products, cloning of GC- AT-rich sequences, module assembly of genetic magenta.
