FIELD: biotechnology.
SUBSTANCE: invention relates to cell biology and biotechnology, namely to methods for generating a population of cells expressing markers characteristic of the pancreatic endoderm line, 60% of which jointly express PDX-1 and NKX6.1, and a method for treating diabetes. Method includes differentiating a population of pluripotent stem cells representing cells from stable lines of human embryonic stem cells H1, H7 or H9 into a population of cells expressing markers characteristic of the definitive endoderm line, and further differentiating said cells into a population of cells expressing markers characteristic of the pancreatic endoderm line in a culture medium supplemented with a protein kinase C activator selected from the group consisting of (2S,5S)-(E,E)-8-(5-(4-(trifluoromethyl)phenyl)-2,4-pentadimoylamino)benzolactam, indolactam V, phorbol-12-myristate-13-acetate and phorbol-12,13-dibutyrate, and also introduction of the obtained cells to a patient suffering from diabetes.
EFFECT: invention allows to obtain cells expressing markers characteristic of the pancreatic endoderm line, 60% of which jointly express PDX-1 and NKX6.1.
