FIELD: cell biology; medicine.

SUBSTANCE: present invention relates to the field of cell biology and medicine, in particular to a method for generation of a population of cells of pancreatic endoderm and a method for the treatment of diabetes, using obtained cells. For the implementation of the specified method for generation of a population of cells expressing markers characteristic for a pancreatic endoderm line, first, the population of cells expressing markers characteristic for a definitive endoderm line is treated with a human fibroblast growth factor. Then, the cell population from the stage a) is treated with cyclopamine-KAAD and retinoic acid. After that, the cell population from the stage b) is treated with a protein kinase C activator. Next, for the treatment of diabetes, a subject is implanted with the resulting cell population. In the resulting population, more than 60% of cells express together PDX-1 and NKX6.1. Markers characteristic for the definitive endoderm line are selected from a group consisting of SOX17, GATA4, HNF3 beta, GSC, CER1, Nodal, FGF8, Brachyury, Mix-like homeobox protein, FGF4, CD48, eomezodermin (EOMES), DKK4, FGF17, GATA6, CXCR4, CKit, CD99, and OTX2. Markers characteristic for the pancreatic endoderm line are selected from a group consisting of PDX1, NKX6.1, HNF1 beta, PTF1 alpha, HNF6, HNF4 alpha, SOX9, HB9, and PROX1.

EFFECT: present invention allows for increase in the efficiency of differentiation of human stem cells to insulin-expressing cells.
