FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry. Population of pancreatic endocrine cells is described, which coexpress NKX6.1 and insulin, and where less than 10% of cells in population express glucagon; population of pancreatic endocrine cells is obtained by differentiation of human pluripotent stem cells, obtained from resistant lines of human embryonic stem cells, in which above differentiation includes first stage of culturing of pluripotent stem cells in medium, containing TGF-receptor agonist-β, selected from group consisting of activin A, activin B, TGFβ-I, TGFβ-II, GDF-8 and GDF-11, in concentration from approximately 2 ng/ml to 100 ng/ml, and additional stage of pancreatic endoderm cell culturing in medium, containing from 20 nm to 500 nm (2S,5S)-(E,E)-8-(5-(4-(trifluoromethyl)phenyl)-2,4-pentadieneoilamino)benzolaktam. Methods of forming these population are also described.
EFFECT: invention extends the range of artificially obtained pancreatic endocrine cells.
