FIELD: biotechnology.
SUBSTANCE: invention relates to a method of obtaining the sequence of the variable domain of a human immunoglobulin heavy chain, a method of producing a fully human heavy chain of immunoglobulin, as well as a method of producing an antibody. These methods are as follows: immunizing a genetically modified mouse with an antigen of interest, where the mouse contains exactly two unrearranged human V immunoglobulin gene segmentsκ and five unrearranged human J immunoglobulin gene segmentsκ operably linked to the mouse immunoglobulin light chain constant region sequence at the endogenous mouse kappa light chain loci, as well as one or more unrearranged human immunoglobulinH gene segments, one or more unrearranged human immunoglobulinH gene segments, and one or more unrearranged human immunoglobulin gene segments human immunoglobulinH segments operably linked to the mouse immunoglobulin heavy chain constant region sequence at endogenous mouse heavy chain loci.
EFFECT: invention is effective for producing fully human antibodies.
