FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry, particularly to a recombinant immunoglobulin light chain nucleic acid comprising at least two unveraging gene segments VL rights and at least one unhearred gene segment JL human functionally related to the immunoglobulin light chain constant region sequence. Moreover, two unprogrammed gene segments VL human are represented by gene segments Vκ1–39 and Vκ3–20 persons, each of which contains at least one histidine codon, which is not coded by corresponding gene segment Vκ1–39 and Vκ3–20 germline of human. Rodent and a method for its preparation, a vector, a rodent cell and tissue containing said nucleic acid are also disclosed. Invention also relates to a method for producing an antibody which exhibits pH-dependent binding with an antigen of interest, involving use of said rodent.
EFFECT: invention enables to efficiently obtain an antibody which exhibits pH-dependent binding with an antigen of interest.
