SUBSTANCE: invention relates to biology and medicine and is a method of preparing a biological sample for examination using scanning electron microscopy, including supravital contrasting, characterized in that a two-step contrasting scheme is used, while as the first contrasting agent an aqueous solution is used based on a mixture of magnesium chloride hexahydrate in a concentration of 1.1–1.8 g/l and chloride hexahydrate of a chemical element of the lanthanide group at a concentration of 13.5–20.2 g/l, and an aqueous solution of lead acetate at a concentration of 82–104 g/l is used as the second contrast agent. In one embodiment, the acidity of the solutions is additionally modified using buffer systems based on acetate anion or caustic soda. In one of the options for storing and fixing the sample after contrasting it is treated with a solution of glutaraldehyde or formalin.

EFFECT: method provides an increase in the contrast of images of biological objects in backscattered electrons with a simultaneous increase in the informativeness of images with the expansion of the list of contrasted ultrastructural elements.
