FIELD: biotechnology.
SUBSTANCE: what is presented is a method for producing target DNA of clinical isolates of urogenital mycoplasmas. Liquid culture sample with concentration of living cells of not less than 106 CFU/ml is pre-treated with a solution containing DNase (100 U) and 10-fold buffer of 100 mM Tris-HCl, 25 mM MgCl2, 1 mM CaCl2 (pH 7.5) (final concentration of buffer in sample should be single). Treatment is carried out for 30±10 minutes at temperature of 37±1 °C. Then enzyme activity is inhibited by heating for 10 minutes at 75±1 °C and targeted target DNA is purified in a magnesium-sorption way.
EFFECT: invention can be used in scientific research practice to obtain a high-quality DNA library for the purpose of subsequent full-genome sequencing.
