FIELD: biotechnology.
SUBSTANCE: in the course of the work, full-genomic DNA sequencing of M. tuberculosis isolates from the collection, in silico polymorphism analysis of virulence genes, detecting correlation between mutations in virulence genes and immunodeficiency of patient, during which mutations in pks15 and re_pgrs54 genes, which are marker for isolates, have been detected, damaging people with immunodeficiencies of various genesis, as well as primers for their detection are developed.
EFFECT: proposed method enables to determine high pathogenicity of mycobacteria for people with low immunity in PCR mode, and, with minimal adaptations, real-time polymerase chain reaction, multilocus PCR with subsequent sequencing, including in MiSeq ("Illumina", USA) and GS Junior ("Roche", Switzerland).
