FIELD: biotechnology.
SUBSTANCE: present invention relates to a method of producing linear or circular DNA and a kit for binding DNA fragments used in said method. Linear or circular DNA is obtained in a reaction solution by linking DNA fragments of two or more types to each other in areas, having homologous base sequences, or in regions having complementary base sequences, in one reaction. Regions having homologous base sequences, or regions having complementary base sequences, are present at the end of the DNA fragment or near it and have length of 20 b. s. or more. Reaction of binding DNA fragments of two or more types is carried out at a temperature in range of 25–48 °C. Reaction solution contains a regenerating enzyme for nucleoside triphosphates or deoxynucleotide triphosphates and a substrate thereof.
EFFECT: method enables to efficiently bind DNA of two or more types using recombinase of the RecA family and exonuclease.
