SUBSTANCE: invention refers to medical equipment, namely to a method for producing tissue-engineered periosteum from cell spheroids for bone defect repair in a subject. In the method, a collagen membrane is placed at the bottom of a culture vessel with a side cover and the collagen membrane is impregnated with a culture medium. Further, a rectangular titanium frame with cross-sectional size of 4 × 4 mm at a frame height of 0.5 mm, the frame presses the edges of the collagen membrane to the bottom of the bottle. A horizontal layer of cell spheroids consisting of aggregated cultured autologous periosteal cells is laid on top of a collagen membrane bounded by the inner walls of the frame in one row. Spheroids are cultivated on the surface of the collagen membrane during maturation of the tissue-engineered structure in a nutrient medium: DMEM or DMEM-F12 450 ml, L-glutamine 292 mg, thrombolysate 50 ml, penicillin 100 units/ml, streptomycin 100 mcg/ml at 100% moisture content and with 5% content of CO2 in the air mixture above the nutrient medium layer inside the culture flask, for 6–7 days with replacement of the culture medium not less than 2 times.
EFFECT: developing a method for producing a bioartificial periosteum containing cultured osteogenic periosteal cells and having a regenerative osteogenic potential when transplanted into a living organism.
