FIELD: medicine; biotechnology.
SUBSTANCE: invention relates to bronchial and lung organoids, methods for preparing them, passaging, cryopreservation, functional analysis of CFTR-channel conductivity. Described are methods for producing bronchial organelles, which include: (a) using a culture medium for producing organoids, containing the following components, %: IMDM 71.8–72.3; F12 (Ham) 23.5–24; B-27 1–2; N2 0.5–1; bovine serum albumin 0.7; thioglycerol 0.004; GlutaMAX 1; L-ascorbic acid 0.2 and primocin 0.2, (b) differentiation of induced human pluripotent stem cells (hIPSC) into definitive endoderm cells using Activin A and CHIR99021, (c) absence of cell sorting at the stage of formation of organelles from NKX2.1 + pulmonary progenitors.
EFFECT: disclosed is a method of producing, culturing and characterizing bronchial and pulmonary organoids from induced pluripotent human stem cells.
