FIELD: biotechnology.
SUBSTANCE: pVEAL3-10H10ch plasmid, CHO-K1-10H10ch recombinant cell line strain, and 10H10ch chimeric antibody against tick-borne encephalitis virus are described. pVEAL3-10H10ch integrative plasmid vector has been created to ensure the expression and secretion of 10H10 antibody against tick-borne encephalitis virus in mammalian cells. The vector has the nucleotide sequence of the following SEQ ID NO: 1, size 6523 b.p. and contains elements according to the physical and genetic map shown in FIG. 1. 10H10ch chimeric antibody against tick-borne encephalitis virus has the amino acid sequences of the heavy chain variable fragments of SEQ ID NO: 2 and light chain SEQ ID NO: 3 and is intended for the creation of therapeutic medicinal products against the tick-borne encephalitis virus. The antibody has a wide cross-activity against other flaviviruses. The invention can be used in genetic engineering, biotechnology and medicine.
EFFECT: obtaining 10H10ch chimeric full-length human antibody against tick-borne encephalitis virus produced by a recombinant strain of CHO-K1-10H10ch Chinese hamster ovary cell line and having broad cross-functional activity against tick-borne encephalitis, Zika, dengue, West Nile, yellow fever viruses.
