SUBSTANCE: invention relates to molecular biology and genetic engineering. Disclosed method of producing a protein of interest, which includes introducing a protein expression vector, which includes a gene fragment containing DNA which codes the protein of interest and a selectable marker gene, as well as transposon Tol1 or Tol2 sequences at both ends of the gene fragment, into a mammalian suspension cell CHO adapted to suspension cultivation, or a PER.C6 cell, or rat myeloma cell YB2/3HL.P2.G11.16Ag.20 (also known as YB2/0), or mouse myeloma cell NS0, adapted to suspension cultivation; integrating the gene fragment inserted between the pair of transposon sequences into the mammalian cell chromosome to obtain a mammalian cell capable of expressing the protein of interest; and suspension cultivation of the mammalian cell; wherein the suspension mammalian cell is capable of expressing the protein of interest. A method of producing a mammalian cell, a corresponding recombinant cell and use of the expression vector are also disclosed.
