SUBSTANCE: invention relates to the field of cell biology and biotechnology, specifically to methods for producing cells expressing markers characteristic of pancreatic endocrine cells, including expression and inducing of PDX1, NKX6.1, and HB9. Methods include differentiation of pluripotent cells obtained without destruction of the embryo, pluripotent cells of non-embryonic origin, embryonic stem cell line H1, embryonic stem cell line H7, embryonic stem cell line H9, inducible pluripotent cells, or reprogrammed pluripotent cells, or definitive endoderm cells derived therefrom, into cells expressing markers characteristic of pancreatic precursor cells of the anterior colon. Resulting cells are differentiated into cells expressing markers characteristic of pancreatic endocrine cells by treatment with at least an ALK5 inhibitor and / or a thyroid hormone selected from triiodothyronine, thyroxine, triiodothyronine analogues, thyroxin analogues or a mixture thereof, and culturing in an air-liquid interface.
EFFECT: invention makes it possible to increase the yield of pancreatic endocrine cells.
