FIELD: medicine.
SUBSTANCE: invention relates to medicine. Disclosed is a method of determining microsatellite instability. Targeted areas are targeted by multiplex PCR, high-efficiency sequencing of the obtained sample of the DNA library, analysis of sequencing results and search of genetic variants with a coating of at least 10 readings and a mutant allele fraction of not less than 0.1 %. Interpretation of detected genetic variants is carried out by determining lengths of fragments. If there are more than 2 versions of lengths for at least 2 and not more than 5 loci, the result is interpreted as low microsatellite instability - MSI-L. If for more than 5 loci there are more than 2 versions of lengths, the result is interpreted as high microsatellite instability.
EFFECT: invention provides high accuracy of sequencing and reduces the probability of erroneous determination of fragment din.
