FIELD: biotechnology; molecular biology; genome editing.
SUBSTANCE: enzyme thermally stable nuclease AflCas9 from Anoxybacillus flavithermus and the application of this enzyme are described. The invention also relates to a nucleic acid encoding a given nuclease, a gene construct, an expression vector that include a given nucleic acid. The invention expands the arsenal of nucleases for genome editing. Due to the wide temperature range of AflCas9, the AflCas9 enzyme is expected to be suitable for editing the genomes of a wide range of organisms that can grow at temperatures from 20 to 60°C.
EFFECT: extended range of activity and stability of AflCa89 will allow it to be used in molecular biology methods that require manipulation of DNA at a temperature of 20-60°C, as well as to be used in complex conditions that require high enzymatic activity and increased stability of the enzyme.
