FIELD: medicine.

SUBSTANCE: invention relates to medicine. The method for obtaining an orthotopic PDX model of squamous cell carcinoma of the human esophagus for the study of radiation therapy in an experiment is that the tumor is excised from the donor’s body. A tumor fragment is isolated without necrosis and vascularization. Next, it is placed in nutrient medium 199 with 1% gentamicin. Anesthesia of Balb/c Nude line mice is carried out. A 27 mm3 tumor fragment is transplanted, dissecting the skin on the neck, the incision begins at the base of the right ear and ends at the base of the left ear. Next, the esophagus is isolated in a blunt way, its adventitial-muscular layer is dissected. Next, a tumor fragment with a volume of 27 mm3 is sewn over the injury using a polypropylene ligature, the surgical wound is sutured with the furrier’s suture using a polypropylene ligature. When the tumor material reaches a volume of 150-250 mm3, xenografts of human esophageal cancer are transplanted this way up to 6 passages.

EFFECT: invention makes it possible to conduct preclinical tests of ionizing radiation with a model characterized by similar morphological features with tumor material obtained from a donor patient.
