FIELD: biotechnology.

SUBSTANCE: plasmid genetic construct pVEAL2-9E2ch-scFv providing expression and secretion of 9E2ch antibody against West Nile virus (WNV) in mammalian cells is proposed, the construct has the nucleotide sequence of the following SEQ ID NO: 1, size 10,498 b.p. and contains elements according to the physical and genetic map shown in FIG. 1, the 9E2-scFv sequence encoding the heavy and light chain variable fragments of the 9E2 antibody connected by a peptide linker. A recombinant strain of the Chinese hamster ovary cell line CHO-K1-9E2ch containing the plasmid genetic construct pVEAL2-9E2ch-scFv and providing expression and secretion of the single-chain antibody 9E2ch against WNV is proposed. The specified strain is deposited under number 324 in the collection of cell cultures of the FBSI SRC VB “Vector” of Rospotrebnadzor. Also the following is proposed: a chimeric single-chain antibody 9E2ch against WNV, which is produced by the specified strain.

EFFECT: invention provides a chimeric single-chain antibody 9E2ch with high affinity for the neonatal FcRn receptor and functional activity against WNV.
