FIELD: medical biotechnology.

SUBSTANCE: disclosed is a method for detecting Mammarenavirus guanaritoense virus by isothermal recombinase polymerase amplification with real-time reverse transcription. Method involves extraction of RNA from biological samples, analysis using reverse transcriptase M-MuLV, specific primers RPA_GTOV_F5'-AAAAGCTTGAAGCCCTCTGTTAAGACCATCGG-3', RPA_GTOV_R5'-CGAAATTTGACAATGGATGAAAAAGTTGTAGTT-3', and fluorescent-labelled DNA probe Exo_GTOV5'-GCAATGCCTCCTTCTGATAAGACAGTTCAGG[FAM-dT]A[dSpacer]A[BHQ1-dT]CAGGTACTTGCCTTC[3'-C3-spacer]-3'. [dSpacer] modification used is the compound dSpacer CE phosphoramidite=5'-O-dimethoxytrityl-1',2'-dideoxyribose-3'-[(2-cyanoethyl)-(N,N-diisopropyl)]-phosphoramidite. Results are interpreted based on the presence or absence of a difference in the fluorescence curve of the sample from the negative control sample which does not contain Mammarenavirus guanaritoense RNA. Result is considered to be positive if the value of the fluorescent signal increases with time and the final value by 30 minutes of analysis exceeds the value in the negative control sample.

EFFECT: method makes it possible to quickly and reliably identify genetic material of Mammarenavirus guanaritoense virus in biological samples and other virus-containing samples.
